Does use or lose leave need to be processed before oct 1, or will leave not processed but taken after or during the Sep/Oct time frame be lost and result in someone using their leave balance minus what they thought they had?

If you have over 60 days, you need to use up what ever amount of days that gets you back down to 60 or less prior to 1 Oct, lest those days over 60 will be gone forever on 1 Oct.

SSG (Join to see) - you have to watch leave taken at the very end of the FY. It is my understanding that the leave (DA31) is charged upon return when S1 forwards it on to Finance. So if you had leave straddling the FYs (example 15Sep-2OCT) there'd be two days from the new FY and the rest from the last. Ensure you get a leave balance before hand. It should show leave.
It is from personal experience that the use lose days are gone in nothing more than a whiff of ozone. Your notification is on your LES, before hand in Use/Lose and then after on the bottom summary.
You need to have this conversation with you BN S1 Shop.

Use or lose need to be taken NLT 30 September. As long as the DA31 shows the leave was taken before 30 September you will not lose the days. Make sure you have a copy of the DA31 in the event your S1 doesn’t process it in time and you lose the days and get charged, that would be your source document for an ETP to get your days credited back to you.
