If a SPC(P) soldier is on assignment to go to Korea and is MTOED for an E5 slot, will they lose that slot if they lose their "P" status?

No, not really. If you were slotted for an E4 position and you were promoted you could lose your spot. E4P is such an easily fluctuating thing that if letting your APFT lapse would cause you to lose your assignment, HRC would be swamped at reassignments.
Also, it's Korea. You could lose a leg and they'd hold your assignment until you came off profile
